The M-82F engine first made it into production La-5s in December 1942, so IIRC it first becomes available in January 1943. For the Kuban phase of career mode, it's enabled by default. 

 

The 20 mm cannon option on the MiG-3 becomes available in 1942. 

 

On the German side, the 37 mm cannons are available for the Ju 87 beginning with Kuban, and the MK 103 is available for the Hs 129 in July 1943. 

 

There are a couple of mods for the Il-2 M42 and M43 (37 mm cannons and PTAB bomblets) that should be date-restricted in career mode, but those have not been implemented yet.
